Neurospora crassa has two large-subunit catalases, CAT-1 and CAT-3. CAT-1 is associated with non-growing cells and accumulates particularly in asexual spores; CAT-3 is associated with growing cells and is induced under different stress conditions. It is our interest to elucidate the structure-function relationships in large-subunit catalases. Here we have determined the CAT-3 crystal structure and compared it with the previously determined CAT-1 structure. Similar to CAT-1, CAT-3 hydrogen peroxide (H2O2) saturation kinetics exhibited two components, consistent with the existence of two active sites: one saturated in the millimolar range and the other in the molar range. In the CAT-1 structure, we found three interesting features related to its unusual kinetics: (a) a constriction in the channel that conveys H2O2 to the active site; (b) a covalent bond between the tyrosine, which forms the fifth coordination bound to the iron of the heme, and a vicinal cysteine; (c) oxidation of the pyrrole ring III to form a cis-hydroxyl group in C5 and a cis-γ-spirolactone in C6. The site of heme oxidation marks the starts of the central channel that communicates to the central cavity and the shortest way products can exit the active site. CAT-3 has a similar constriction in its major channel, which could function as a gating system regulated by the H2O2 concentration before the gate. CAT-3 functional tyrosine is not covalently bonded, but has instead the electron relay mechanism described for the human catalase to divert electrons from it. Pyrrole ring III in CAT-3 is not oxidized as it is in other large-subunit catalases whose structure has been determined. Different in CAT-3 from these enzymes is an occupied central cavity. Results presented here indicate that CAT-3 and CAT-1 enzymes represent a functional group of catalases with distinctive structural characteristics that determine similar kinetics.  相似文献

We describe a few mathematical tools which allow to investigate whether air-water interfaces exist(under prescribedconditions)and are mechanically stable and temporally persistent.In terms of physics,air-water interfaces are governed by theYoung-Laplace equation.Mathematically they are surfaces of constant mean curvature which represent solutions of a nonlinearelliptic partial differential equation.Although explicit solutions of this equation can be obtained only in very special cases,it is-under moderately special circumstances-possible to establish the existence of a solution without actually solving thedifferential equation.We also derive criteria for mechanical stability and temporal persistence of an air layer.Furthermorewe calculate the lifetime of a non-persistent air layer.Finally,we apply these tools to two examples which exhibit the symmetriesof 2D lattices.These examples can be viewed as abstractions of the biological model represented by the aquatic fernSalvinia.  相似文献

Strain differences in delay discounting using inbred rats   总被引:1,自引:0,他引:1  
A heightened aversion to delayed rewards is associated with substance abuse and numerous other neuropsychiatric disorders. Many of these disorders are heritable, raising the possibility that delay aversion may also have a significant genetic or heritable component. To examine this possibility, we compared delay discounting in six inbred strains of rats (Brown Norway, Copenhagen, Lewis, Fischer, Noble and Wistar Furth) using the adjusting amount procedure, which provides a measure of the subjective value of delayed rewards. The subjective value of rewards decreased as the delay to receipt increased for all strains. However, a main effect of strain and a strain × delay interaction indicated that some strains were more sensitive to the imposition of delays than others. Fitting a hyperbolic discount equation showed significant strain differences in sensitivity to delay ( k ). These data indicate that there are significant strain differences in delay discounting. All strains strongly preferred the 10% sucrose solution (the reinforcer in the delay discounting task) over water and the amount of sucrose consumed was correlated with sensitivity to delay. Locomotor activity was not correlated with delay discounting behavior. Additional research will be required to disentangle genetic influences from maternal effects and to determine how these factors influence the underlying association between heightened delay discounting and neuropsychiatric disorders.  相似文献

AimsStudies have shown that the acute administration of venlafaxine elicits an antidepressant-like effect in the mouse forced swim test (FST) by a mechanism dependent on the l-arginine–nitric oxide (NO)–cyclic guanosine monophosphate (cGMP) pathway. Because it has been reported that NO activates different types of potassium (K+) channels in the brain, this study investigated the involvement of K+ channels in the antidepressant-like effect of venlafaxine in the mouse FST.Main methodsMale adult Swiss mice were pretreated with different K+ channel inhibitors or openers 15 min before venlafaxine administration. After 30 min, the open-field test (OFT) and FST were carried out.Key findingsIntracerebroventricular (i.c.v.) pretreatment of mice with subeffective doses of tetraethylammonium (TEA, a non-specific inhibitor of K+ channels, 25 pg/site), glibenclamide (an ATP-sensitive K+ channel inhibitor, 0.5 pg/site), charybdotoxin (a large- and intermediate-conductance calcium-activated K+ channel inhibitor, 25 pg/site) or apamin (a small-conductance calcium-activated K+ channel inhibitor, 10 pg/site) was able to potentiate the action of a subeffective dose of venlafaxine (2 mg/kg, i.p.). Moreover, the reduction in the immobility time elicited by an effective dose of venlafaxine (8 mg/kg, i.p.) in the FST was prevented by the pretreatment of mice with the K+ channel openers cromakalim (10 µg/site, i.c.v.) and minoxidil (10 µg/site, i.c.v.). The drugs used in this study did not produce any change in locomotor activity.SignificanceThe results demonstrate that the neuromodulatory effects of venlafaxine, via the inhibition of K+ channels, are possibly involved in its anti-immobility activity in the mouse FST.  相似文献

Choosing an effective experimental design generally results from hypotheses derived from prior knowledge or experimentation. In drug discovery, the characterisation of the precise modes of action (MoA) and of unwanted off-target effects of novel molecularly targeted compounds is of highest relevance. Recent approaches for identification of MoA have employed various techniques for modeling of well defined signaling pathways including structural information, changes in phenotypic behavior of cells and gene expression patterns after drug treatment. However, efficient approaches focusing on proteome wide data for the identification of MoA including interference with mutations are underrepresented. As mutations are key drivers of drug resistance in molecularly targeted tumor therapies, efficient analysis and modeling of downstream effects of mutations on drug MoA is a key to efficient development of improved targeted anti-cancer drugs. Here we present a combination of a global proteome analysis, reengineering of network models and integration of apoptosis data used to infer the mode-of-action of various tyrosine kinase inhibitors (TKIs) in chronic myeloid leukemia (CML) cell lines expressing wild type as well as TKI resistance conferring mutants of BCR-ABL. The inferred network models provide a tool to predict the main MoA of drugs as well as to grouping of drugs with known similar kinase inhibitory activity patterns in comparison to drugs with an additional MoA. We believe that our direct network reconstruction approach, demonstrated on proteomics data, can provide a complementary method to the established network reconstruction approaches for the preclinical modeling of the MoA of various types of targeted drugs in cancer treatment. Hence it may contribute to the more precise prediction of clinically relevant on- and off-target effects of TKIs.  相似文献
